The Effect of Various Storage Conditions on the Measurement of Aminotransferases



Abstract
Aspartate aminotransferase (AST) and alanine aminotransferase(ALT) enzymes are important, because there are many patients suffered from liver disease in Korea. Because the test of anti-HCV is not sufficient to screen for Non-A Non-B(NANB)
hepatitis,
ALT is still a useful surrogate marker of NANB hepatitis. So accurate analysis of these aminotransferases is essential in clinical laboratory. The biochemical tests cannot be procedured daily in small military hospital due to some problems. So
many
preanalytical errors may influence the results of these enzymes. In this study, pre-analytical errors on these enzymes were evaluated. 50 mililiters from 10 healthy young volunteers were sampled, and dispensed into 10 glass tubes. Five tubes were
stored
at room temperature (RT) and the others at 4¡É. Each five were conditioned as 1) whole blood, centrifuged; 2) whole blood, centrifuged, capped; 3) whole blood, not centrifuged; 4) whole blood, not centrifuged, capped; 5) separated serum. Each one
was
analyzed at the 0, lst, 3rd and 5th day. Another 10 voluntters were sampled, and analyzed at the 0.7th , 10th 14th day at the same conditions.
AST was increased significantly at the 3 rd day when un-capped samples were stored at room temperature. It was increased at the 7th day in capped condition. If samples were stored in refrigerator, AST was significnatly increased at the 10th day
in
un-capped condition, at the 14th day in capped condition respectively.
Alt was significantly increased at the 5th day in un-capped condition, at the 7th day in capped condition respectively. If samples were stored at refrigerator, ALT was significantly increased at the 14th day. AST and Alt of the sera stored at
room
temperature were decreased about 3% per day. If the sera wee stored in refrigerator, aminotranferases were relatively stable during 7 days.
